This application relates to antibody scaffolds for nanobody humanization.Shark variable domain of new antigen receptor (vNAR) has emerged as an up-rising star in novel antibody-based therapies. Multiple studies have reported their vNAR with potential disease treatment function, such as treating the tumor, malaria, SARS-CovID19, Ebola hemorrhagic fever, CNS diseases, and inflammation. Although much effort has been exerted in the significant concerns, Achilles heels remain to advance these emerged milestones to safe clinical utilization, mainly contributed by immunogenicity that these potent novel therapeutic vNARs may trigger during long-term administration. Almost all the above-described studies fixed their minds on highlighting the antigen-antibody reactivity-based molecular and cellular experiment outputs while losing their sight of (or shunned) the immune response vNAR could cause as an exogenous substrate. Hence investigating the immunogenicity and its solution of target-specific shark vNAR will be the gap-filling work.
